Single-Cell Analysis of the Aging Human Periovulatory Follicle Reveals Granulosa Cell Dysfunction and Immune Dysregulation
Listed in NCBI GEO
Advanced maternal age is a major cause of declining female fertility, yet how aging reshapes the cellular landscape of the human periovulatory follicle at single-cell resolution remains incompletely defined.
Description
We performed single-cell RNA sequencing (10x Genomics Chromium 3' v3) of human follicular aspirates collected 36 hours after hCG administration during in vitro fertilization for male-factor infertility, comparing women of optimal maternal age (OMA; 21-29 years) and advanced maternal age (AMA; >=41 years).
Across 19 annotated follicular and immune cell types, advanced maternal age was associated with two concurrent, compartment-specific programs: a coordinated downregulation of oxidative phosphorylation and of ovulatory and granulosa-oocyte communication genes in the granulosa cell lineage, and an upregulation of TNFa-NFkB / inflammatory response signaling with dysregulated neutrophil expansion in the immune compartment, together with a global weakening of intercellular communication.

Canonical senescence signatures were not increased in the granulosa cell lineage.
