Simultaneous analysis of leukemic mutations and transcriptomes at the single-cell level

Single-cell RNA-sequencing (scRNA-seq) enables detailed profiling of transcriptional heterogeneity in leukemic cells, but genetic variants are typically assessed by using separate assays, limiting the ability to link the genotype with the transcriptional state of a cell.

Description

We developed a method that enables the simultaneous genotyping and transcriptomic profiling of leukemic cells. We repurposed the intermediate barcoded full-length cDNA amplicons that are generated during the standard 3' 10X Genomics scRNA-seq workflow to selectively enrich for the transcripts of interest and subject them to PacBio long-read sequencing.

This approach enables the integrated analysis of somatic mutations and gene expression in the same cells.
