Census of clustered protocaherins in human semi-guided organoids and 1st trimester embryo at single-cell resolution [PCDH_human_embryo]

Clustered protocadherins are cell adhesion proteins expressed at the cell surface of neurons and other brain cells.

Description

The establishment of their expression patterns follows rules that are distinct during development compared to adult cells. It remains unknown how these rules are determined and whether are followed in a cell-type specific manner.

This study provides the first census of clustered protocadherins in hiPSC-derived organoids at single-cell resolution using 5' scRNA-seq to capture the distinct region of each member of the clustered protocadherin family and scATAC-seq to characterize the chromatin structure acrosss the locus. A 3' scRNA-seq dataset is used to integrate the 5' scRNA-seq and scATAC-seq datasets by combining the 5' and 3' datasets and leveraging the cellular barcodes in the 3' dataset as anchors for the scATAC-seq dataset, obtained by 3' scRNA/ATAC-seq.
