CREBBP Loss Promotes Immune Evasion by Silencing STING and MHC I Expression in Small Cell Lung Cancer [RNA-Seq]

Most patients with small cell lung cancer (SCLC) exhibit primary resistance to immunotherapy, yet the molecular mechanisms underlying this resistance remain poorly understood.

Description

Here we identify that CREBBP loss, a frequent genetic alteration in SCLC, induces an immune-cold tumor microenvironment in Rb1/Trp53-driven mouse models. Mechanistically, Crebbp loss epigenetically suppresses Sting and MHC I expression by decreasing H3K27 and H3K18 acetylation.
